Flow Cytometry

Flow Cytometry

Flow Cytometry is a laser-based technology used to cell counting, cell sorting, biomarker detection and protein engineering. Analyzes characteristics of particles of fluid as it passes through lasers. Most commonly utilized for diagnostic testing for leukemias, lymphomas, myeloproliferative disease, myelomas and myelodysplastic syndrome.

VPL utilizes 10-Color Flow Cytometry, up to 43 antibodies.

Specimen Requirements

Bone marrow aspirate: 1-3 mL in green-top (sodium heparin) tube

Peripheral blood: 3-10 mL in green-top (sodium heparin) tube

Fresh bone marrow core: 1-2 cm in RPMI

Fresh Tissue / Lymph Nodes in RPMI

Body Fluids: Equal part specimen volume to RPMI

CSF in sterile container

Stability

Blood / Bone Marrow – stability 72 hours.  Please submit within 24 hours whenever possible.

Tissue / Fluids – submit within 24 hours for optimal specimen stability

Storage and Transportation

Blood and bone marrow at room temperature.

Tissue, body fluids and FNA : Refrigerate specimens. Cold pack for transport. DO NOT FREEZE.

Submit specimens within 24 hours.

Morphology

Specimen Requirements

A bone marrow core biopsy and BM aspirate clot, each in a separate fixative container.

Air-dried and/or stained slides to include 3 fresh bedside aspirate smears and biopsy touch imprints/preps in plastic slide case.

Peripheral blood in green-top (sodium heparin) tube preferred.

NOTE: For a bone marrow evaluation also include peripheral blood

Stability

Blood / Bone Marrow – stability 72 hours

Storage and Transportation

Ambient temperature; however OK to use cold pack for transport.

Specimen Requirements

Bone marrow aspirate: 1-3 mL in green-top (sodium heparin) tube

Peripheral blood: 5-10 mL in green-top (sodium heparin) tube

Fresh bone marrow core: 1-2 cm in RPMI

Fresh Tissue / Lymph Nodes in RPMI

Voided Urine: Minimum 33 mL, if exceeds 60 mL poor off. 2 parts urine to 1 part preservative. Total fluid volume not to exceed 90mL.

*Urine kits for FISH Urovysion provided by VPL.

Blood / Bone Marrow – stability 72 hours

Storage and Transportation

Molecular

Peripheral blood:   1-3 mL in lavender-top (EDTA) tube

Bone marrow: 1-3 mL in lavender-top (EDTA) tube

FFPE Tissue: Paraffin block or 6 unstained slides cut at 4 μm or 5 unstained slides cut at 4 μm and 1 H & E (preferable).

Stability

Blood / Bone Marrow – stability 72 hours

Urovysion by FISH
